rs6025 is the F5 Arg506Gln missense variant, known as Factor V Leiden, which destroys one of three cleavage sites that activated protein C (APC) uses to inactivate clotting factor Va. The mutated factor Va is degraded far more slowly, producing APC resistance and a hypercoagulable state that raises lifetime risk of venous thromboembolism (DVT/PE) roughly three- to eightfold in heterozygotes. It is the primary marker behind the Blood-clot risk (hereditary thrombophilia) trait.
