F5 encodes coagulation factor V, a plasma glycoprotein that serves as a cofactor for factor Xa in the prothrombinase complex that converts prothrombin to thrombin, and is itself inactivated by activated protein C to limit clotting. The variant rs6025 (Factor V Leiden) makes factor V resistant to inactivation by activated protein C, producing a hypercoagulable state and making it the most common inherited risk factor for venous blood clots.
