rs225014 is the DIO2 Thr92Ala missense variant: the common allele encodes threonine and the minor allele encodes alanine at position 92 of type 2 iodothyronine deiodinase (DIO2), the enzyme that converts inactive thyroid hormone T4 into active T3. The Ala92 variant reduces deiodinase-2 enzymatic activity and lowers T3 output relative to T4 in thyroid-deficient patients, and has been linked to insulin resistance in some populations. It is used alongside DIO1 rs2235544 as a marker of T4-to-T3 conversion efficiency.
