MC4R

MC4R (melanocortin 4 receptor) encodes a G-protein-coupled receptor expressed mainly in the hypothalamus that responds to alpha-MSH and AgRP signaling to regulate satiety, energy expenditure, and food intake. Common variants near MC4R, including rs17782313, are among the most robustly replicated GWAS hits for body weight, BMI, and appetite tendency in the general population, distinct from rare monogenic MC4R mutations that cause severe early-onset obesity.
