DPYD
DPYD encodes dihydropyrimidine dehydrogenase, the rate-limiting enzyme in the catabolism of uracil and thymine that also degrades fluoropyrimidine chemotherapy drugs such as 5-fluorouracil and capecitabine. Loss-of-function DPYD variants reduce enzyme activity, causing these drugs to accumulate and raising the risk of severe, sometimes life-threatening toxicity, which is why DPYD genotype is used clinically to guide fluoropyrimidine dosing.
