COL5A1 encodes the alpha-1 chain of type V collagen, a minor fibrillar collagen that nucleates and regulates the diameter of type I collagen fibrils in tendons, ligaments, and skin. A 3' UTR variant tagged by rs12722 is associated with tendon and ligament stiffness, and has been linked in multiple studies to susceptibility to soft-tissue injuries (e.g., Achilles tendinopathy) and exercise-induced muscle soreness.
