rs4680 is the COMT Val158Met missense variant in catechol-O-methyltransferase, the enzyme that degrades synaptic dopamine, norepinephrine, and catechol estrogens. The Met allele reduces COMT enzymatic activity roughly three- to four-fold compared with Val, slowing catecholamine clearance and raising prefrontal dopamine tone. This activity difference underlies the Dopamine & catecholamines trait, covering dopamine signaling, stress reactivity, and estrogen-pathway processing.
