rs307355 is a C>T single-nucleotide variant located about 1.5 kb upstream of TAS1R3, in the gene's promoter region, rather than a coding change. The T allele reduces TAS1R3 promoter transcriptional activity relative to the C allele, and behavioral testing links T-allele carriers to reduced ability to discriminate sucrose sweetness intensity, explaining a substantial share of population variation in sweet-taste sensitivity. It is used together with TAS2R16 rs846672 to characterize combined sweet and bitter taste sensitivity.
