VKORC1
VKORC1 encodes vitamin K epoxide reductase complex subunit 1, an endoplasmic reticulum enzyme in the liver that recycles oxidized vitamin K back to its active reduced form, which is required to gamma-carboxylate and activate clotting factors II, VII, IX, and X. VKORC1 is the direct molecular target of warfarin, and promoter and coding variants in this gene are a major determinant of warfarin dose requirement and bleeding sensitivity.
