PDE3B lies on chromosome 11p15.2 directly adjacent to CYP2R1, the enzyme that hydroxylates vitamin D in the liver, and this shared chromosomal locus shows genome-wide association with circulating 25-hydroxyvitamin D levels, which is why PDE3B is flagged in vitamin D genetic analyses even though CYP2R1 is thought to drive the underlying biology.
