ADH1B encodes the beta subunit of class I alcohol dehydrogenase, the primary liver enzyme that catalyzes the first step of ethanol metabolism by oxidizing alcohol into acetaldehyde. The rs1229984 variant (His47Arg, the ADH1B*2 allele) produces a markedly more catalytically active enzyme that clears ethanol to acetaldehyde much faster than the ancestral form, a difference that speeds subjective alcohol effects and is strongly protective against heavy drinking and alcohol use disorder.
